FORMULASI KAPSUL EKSTRAK KENTAL ETANOL 96% TERIPANG KELING (Holothuria atra) dan UJI AKTIVITAS ANTIBAKTERI

Abstract


ABSTRAK

Organisme hidup tidak terkecuali biota laut menghasilkan berbagai produk alami yang terdiri dari atas metabolit primer dan metabolit sekunder.Beberapa kandungan bioaktif pada biota laut telah terbukti secara ilmiah memiliki aktifitas atibakteri, antikoagulan, antifungi, antiinflamasi, antimalaria dan antivirus.Salah satu biota laut yang masih terus diteliti sebagai bahan obat adalah teripang.Teripang berpotensi untuk menghasilkan metabolit sekunder, sehingga dapat dimanfaatkan substansi aktif dalam bidang obat-obatan yang belum banyak diteliti khususnya di Indonesia. Penelitian ini bertujuan untuk mendapatkan konsentrasi hambat minimum ekstrak teripang keling (Holothuria atra) dan untuk membuat formulasi kapsul ekstrak kental teripang keling (Holothuria atra)  dengan  berbagai konsentrasi. Hasil penelitian menunjukkan bahwa ekstrak kental teripang dengan konsentrasi 100µ g menghambat pertumbuhan bakteri gram positif yaitu bakteri Bacillus subtilis, 12 mm dan Staphylococcus aureus 11mmdan Gram negatif yaitu Escherichia coli 12 mm,Vibrio cholera 9mm. Berdasarkan hasil  zona hambat bakteri, formulasi kapsul ekstrak teripang menggunakan kosentrasi 100µg, 250µg dan 500µ g. Berdasarkan hasil evaluasi laju alir, sudut istirahat, keseragaman bobot, waktu hancur dan kadar air pada penyimpanan selama 5 minggu formula A, B dan C menunjukkan ketiga formula memenuhi persyaratan sediaan kapsul.

ABSTRACT

A living organism include marine lifes produce a variety of natural products consisting of upper primary metabolites and secondary metabolites. Some bioactive compounds in marine lifes have been scientifically proven have antibacterial, anticoagulant, antifungal, anti-inflammatory, antimalarial and antiviral activities. One of marine lifes that is still evaluated as a drug ingredient is sea cucumber. Sea cucumber has ability to produce secondary metabolites and it can be used as an active substance in the field of medicine that has not been extensively studied, especially in  Indonesia. The  purposes  of this  study were  to  obtain the minimum inhibitory concentration of sea cucumber extract (Holothuria atra) and make sea cucumber extract in capsule formulation with various concentrations. The results showed that the thick extract of sea cucumber with  100 μg concentration inhibiting the growth of gram-positive bacteria, Bacillus subtilis and Staphylococcus aureus by successively are 12 mm and 11 mm while Gram-negative bacteria Escherichia coli and Vibrio cholerae were 12 mm and 9 mm. Based on the results of bacterial inhibition zone, sea cucumber extract capsule formulation use a concentration of 100 μg, 250 μg and 500 μg. Based on the evaluation of flow rate, angle of rest, weight uniformity, disintegration time, and moisture content on saving for 5 weeks it can be concluded that all  of formulas have requirements ascapsules.
